In my experience yes. I was the author of such a thread on my realm forums (
WoW Forums -> Howto: Reduce your Latency).
I run Vista Home Premium and the fix still makes around 100-150ms difference to my ping as measured by recount's realtime graph. I done hours of testing on this subject and it makes a big difference.
