There is 5th gliph. Glyph of Death Strike.
After any killing blow u can use DS to free 30-60k dmg (and heal yourself of course).
It works wery well on 3rd boss for example. After killing mob in dark room u can do free dmg (usually runes not refreshed so fast) to next add and heal yourself through shadowbolts damage.
